This Thursday May 27th, at 3pm, Paweł Segit, a PhD student from the Laboratory of of Molecular Neurobiology led by Prof. Bożena Kamińska, will give a lecture entitled: Heterogeneity of human glioma-associated brain macrophages at single-cell resolution.

Abstract

Glioblastoma is the most aggressive primary brain tumor that exhibits massive infiltration of microglia (brain resident immune cells) and peripheral macrophages. Glioma-derived factors drive accumulation and polarization of these myeloid cells to glioma-associated microglia and macrophages (GAMs), which exhibit immunosuppressive activity, promote tumor invasion and angiogenesis. Several studies have shown that GAMs are not a uniform population and might consist of distinct phenotypic or functional subpopulations. To bring new insights into this field, we decided to perform single-cell RNA sequencing of CD11b+ cells isolated from samples collected intraoperatively from patients diagnosed with glioblastoma. Single-cell RNA sequencing offers remarkable possibility to increase resolution of transcriptional studies and to obtain gene expression profiles from actual single cells. In the next step we performed unbiased clustering of collected cells, based on the acquired gene expression profiles, that resulted in identification of multiple CD11b+ subpopulations. Following work focused on identification of markers that allow to separate and functionally characterize these subpopulations. Results of our analyses present biologically meaningful transcriptomic differences among GAMs, contribute to better understanding of characteristics and heterogeneity of glioblastoma microenvironment, and subsequently might help in improving existing treatment procedures.
