On the next seminar of the Laboratory of Neuroinformatics Dirk Schubert (Donders Institute for Brain, Cognition and Behaviour Radboud University Nijmegen Medical Centre) will give a talk: "The Ying & Yang of neuronal network activity - Investigating E/I balance in health and disease"

 

Abstract:

"Alterations of the interplay of GABAergic inhibitory and glutamatergic excitatory signaling in neuronal networks, the so-called E/I (dys)balance, are implied to represent a mechanistic commonality in neurodevelopmental disorders. In this presentation, after defining what E/I (dys)balance conceptually reflects, we will discuss a cross-species (human-rodent) approach that allows assessing the E/I balance qualitatively and quantitatively in the neuronal circuitry.  Reductionistic rodent and human iPSC derived neuronal in vitro approaches, including 2-dimensional neuronal cultures growing on multi electrode arrays, allow identifying main molecular and cellular mechanisms that contribute to altered E/I balance during early network formation and maturation. Rodent approaches, such as behavioral assessments and whole brain fluorescent imaging, allow revealing the consequences of such E/I balance alterations during the early brain development on the mature brain organization and function. For example, in Kleefstra syndrome, a genetic disorder associated with intellectual disability, we show that impaired E/I balance during early network formation may lead to very complex brain region specific changes in network organization which eventually result in prominent behavioral phenotypes."
